Effect of Deep Sea Water containing Chitosan Oligosaccharides on Fatty Acids Synthesis, ¥â-oxidation, and AMPK Activation

Abstract

|
To determine effect of deep sea water (DSW) containing chitosan oligosaccharides (COS) on fatty acids synthesis and ¥â-oxidation, we performed GPDH activity assay and RT-PCR in differentiated or mature 3T3-L1 adipocytes with the treatment of DSW or DSW containing COS. In differentiated 3T3-L1 adipocytes, both DSW and DSW containing COS treatment inhibited GPDH activity and the expressions of genes related to fatty acids synthesis such as SREBP 1c, FAS, and aP2. On the other hands, the expressions of genes related to ¥â-oxidation such as CPT 1, MCAD, ACO, and UCP 2 were increased in mature 3T3 L1 adipocytes. Furthermore, the phosphorylation of AMPK also was increased by DSW and DSW containing COS treatment in both differentiated and mature 3T3-L1 adipocytes. Taken together, these data suggest that DSW containing COS is more effective than DSW alone in inhibitory effect on fatty acids synthesis and stimulatory effect on ¥â-oxidation and AMPK activation.
